"We first pretreated THP-1 macrophages transfected with Ctrl siRNA or GMFG siRNA with 10 ng/ml LPS for 24 h and subsequently challenged them with 100 ng/ml LPS for 24 h. Consistent with published reports describing LPS induced endotoxin tolerance, pretreatment with low-dose LPS nearly completely inhibited TNF-alpha, IL-6, and IFN-beta production after secondary LPS challenge in THP-1 macrophages transfected with Ctrl siRNA (XREF_FIG)."
